EPR Detection of the Superoxide Free Radical with the Nitrone Spin Traps DMPO and BMPO

چکیده

acterizing free radicals in chemistry, biology and medicine, is detection by EPR spectroscopy. However, due to their high reactivity and short half-lives, direct EPR detection of many free radicals (e.g., superoxide, hydroxyl radical, alkyl radicals, etc.) is virtually impossible in solution at room temperature. Spin trapping is a technique developed in the late 1960s in which a nitrone or nitroso compound reacts with a target free radical to form a stable and identifiable free radical that is detected by EPR spectroscopy. The spin trapping technique involves the addition of the reactive free radical across the double bond of a diamagnetic ‘‘spin trap’’ to form a much more stable free radical (a ‘‘radical adduct’’) which can then be measured with EPR: other nitrone spin traps. It is very redox inert. DMPO forms radical adducts with O-, C-, N-, S-centered radicals that have very distinguishable EPR spectra. This allows the researcher to identify the type of free radical that was formed in a given reaction. This is not the case for other spin traps such as α-phenyl-N-tert-butylnitrone (PBN) where the EPR spectra for the radical adducts are nearly identical regardless of the radical trapped. The identity of a DMPO radical adduct is then identified using references from a web search of related scientific literature.

Increased flux of free radicals in cells subjected to hyperthermia: detection by electron paramagnetic resonance spin trapping.

It has been hypothesized that hyperthermia promotes oxygen-centered free radical formation in cells; however, to date there is no direct evidence of this heat-induced increase in oxygen free radical flux. Using electron paramagnetic resonance (EPR) spin trapping, we sought direct evidence for free radical generation during hyperthermia in intact, functioning cells. Rat intestinal epithelial cel...

In vitro free radical metabolism of phenolphthalein by peroxidases.

Phenolphthalein, a widely used laxative, is the active ingredient in more than a dozen commercial nonprescription formulations. Fast-flow EPR studies of the reaction of phenolphthalein with horseradish peroxidase (HRP) and hydrogen peroxide permit the direct detection of two free radicals. One has EPR parameters characteristic of phenoxyl radicals. The other has a broad unresolved spectrum, pos...

Detection and imaging of the free radical DNA in cells—Site-specific radical formation induced by Fenton chemistry and its repair in cellular DNA as seen by electron spin resonance, immuno-spin trapping and confocal microscopy

Oxidative stress-related damage to the DNA macromolecule produces lesions that are implicated in various diseases. To understand damage to DNA, it is important to study the free radical reactions causing the damage. Measurement of DNA damage has been a matter of debate as most of the available methods measure the end product of a sequence of events and provide limited information on the initial...

Imaging free radicals in organelles, cells, tissue, and in vivo with immuno-spin trapping

The accurate and sensitive detection of biological free radicals in a reliable manner is required to define the mechanistic roles of such species in biochemistry, medicine and toxicology. Most of the techniques currently available are either not appropriate to detect free radicals in cells and tissues due to sensitivity limitations (electron spin resonance, ESR) or subject to artifacts that mak...

Synthesis and EPR Evaluation of the Nitrone PBN-[tert-13C] for Spin Trapping Competition

Electron Paramagnetic Resonance, Spin Trapping, Free Radicals N-[tert-13C]Butyl C-phenyl nitrone (PBN-[tert-13C]) has been synthesized for an EPR spin trapping competition study. The newly synthesized PBN-[tert-13C] shows different 13C-hyperfine splitting constants (a13C) when it traps free radicals as compared to another 13C-labeled PBN analogue, N-tert-butyl C-phenyl [nitronyl-13C]nitrone (PB...
